How does DB2 and MySQL differ? Is DB2 expess C and MySQL are similar in functionality? Please direct me to some link if there are some discussion on this.

Google is your friend. Googling db2 mysql returned about 557,000 hits, and several of the hits on the first page addressed your questions.

One of the main difference is that MySQL is not supported by zOS but on the other hand, DB2 has its sibling working for zOS. In fact it all evolved from zOS. Both of them are relational database system (just like Oracle). MySQL is widely used Open Source RDBMS while DB2 is common in IBM hosted environments.
DB2 Express-C is basic and free edition but I believe even with that it's more powerful than MySQL but I've not done a comparative study of them to set a bench mark. By the way, in the past, MySQL was an Open Source project, but now it is part of Oracle. And Oracle has finished the support to many of the Open Source projects that came from Sun and as such you never know the future while IBM has released DB2-Express C for "free".
